Use and control of cephalosporins.

چکیده

It is now over 20 years since Professor Brotzu recovered the mould Cephalosporium acremonium from Sardinian sewage and found that it produced antibiotic material. Figure 1 represents a simplified cephalosporin family tree. The first antibiotic to be isolated from the mould was cephalosporin N, an acyl derivative of 6-aminopenicillanic acid, susceptible to the action of stapylococcal penicillinase and much less active against Gram-positive bacteria than benzyl penicillin. Cephalosporin P consists of one major component and at least four minor components: its major component is a steroid related chemically to, but considerably less active than, fusidic acid. The most important antibiotic elaborated by the cephalosporium mould is cephalosporin C, but it is only produced in minute quantities. It is from cephalosporin C that the semi-synthetic cephalosporins have been derived. Several hundred analogues have been produced by substitution of different chemical groups at the 7 and 3 position (R1 and R2 in the diagram) of the cephalosporin nucleus and have been investigated for antimicrobial activity. Four cephalosporins have been extensively studied in various parts of the world-cephaloridine, cephalothin, cephalexin, and cephaloglycine-and of these the first three drugs are now available commercially in Britain and it is with them that this paper is concerned. Cephaloridine has been available since 1964 and cephalothin and cephalexin much more recently.

Pixel selection by successive projections algorithm method in multivariate image analysis for a QSAR study of antimicrobial activity for cephalosporins and design new cephalosporins

Thirty-one Cephalosporin compounds were modeled using the multivariate image analysis and applied to the quantitative structure activity relationship (MIA-QSAR) approach. The acid dissociation constants (pKa) of cephalosporins play a fundamental role in the mechanism of activity of cephalosporins. The antimicrobial activity of cephalosporins was related to their first pKa by different models. B...

Resistance of Shigella strains to extended-spectrum cephalosporins in Isfahan province

Background: The aim of this study was to determine the serotypes and antimicrobial susceptibility of Shigella spp. in Isfahan, (Iran) from 2010 to 2015.   Methods: This retrospective study was conducted on Shigella isolates in four tertiary care hospitals. The process of bacterial isolation and determination of susceptibility was performed by standard microbiological guidelines. The pat...

Use of Hypoprothrombinemia-Inducing Cephalosporins and the Risk of Hemorrhagic Events: A Nationwide Nested Case-Control Study

OBJECTIVE Existing data regarding the risk of hemorrhagic events associated with exposure to hypoprothrombinemia-inducing cephalosporins are limited by the small sample size. This population-based study aimed to examine the association between exposure to hypoprothrombinemia-inducing cephalosporins and hemorrhagic events using National Health Insurance Research Database in Taiwan. DESIGN A na...

Hospital-based strategies for combating resistance.

Selective pressures generated by the indiscriminate use of beta-lactam antibiotics have resulted in increased bacterial resistance across all beta-lactams classes. In particular, the use of third-generation cephalosporins has been associated with the emergence of extended-spectrum beta-lactamase-producing and AmpC beta-lactamase-producing Enterobacteriaceae and vancomycin-resistant enterococci....

Minimum Inhibitory Concentration of Cefotaxime by E-test Method on Klebsiella in Gorgan

Abstract Background and objectives: the increasing use of antibiotics, especially the third generation cephalosporins, is an important factor in the spread of antibiotic resistance in bacteria. The main reason for the development of resistance phenotype such as Extended Spectrum Beta Lactamas (ESBL) is the extensive use of broad-spectrum cephalosporins. In phenotypic survey, the Phenotyping c...
